PARP Inhibitor | Cancer Research News

PARP Inhibitor are substances that that have an action on cancer cells altering their ability to grow and replicate DNA.  According to some studies it should make chemotherapy more effective.  The use of PARP inhibitors has been discussed greatly over the last years, and also its application with chemotherapy.

On recent news, we get the information that PARP inhibitors show specificity for BRCA tumors. From Medpage Today we have the following info:

An investigational cancer therapy demonstrated significant antitumor activity in patients with BRCA1/2 mutations, according to the investigators in the most recent string of favorable studies. [...]only mutation carriers had responses to the oral inhibitor of poly(adenosine diphosphate [ADP]-ribose) polymerase (PARP). However, the agent demonstrated activity in patients with breast, ovarian, and prostate cancer.

Authors of the study said:

“This tumor-specific defect can be exploited by using PARP inhibitors to induce selective tumor cytotoxicity, sparing normal cells,PARP inhibition in these tumor cells with deficient homologous-recombination repair generates unrepaired DNA single-strand breaks that are likely to cause the accumulation of DNA double-strand breaks and collapsed replication forks.”
